Article Snippet: Then at three time points—1 h, 3 h, or 6 h after infection, the cells were fixed with 3.8% formaldehyde for 10 min at RT, then washed in PBS for 5 min and treated with 0.2% Triton X-100 for 10 min, after which cells were again washed once and incubated with 5% goat serum in PBS (blocking buffer) for 1 h. Then cells were incubated with SDOW17 (FITC conjugated anti-PRRSV N) monoclonal Ab at 1:80 dilution, together with either anti-EEA1 Ab (Santa Cruz Biotechnology, sc-365652, Dallas, TX, USA) or anti-Lamp-1 Ab (Santa Cruz Biotechnology, sc-19992) conjugated to Alexa Fluor 647.
